Yes its first in last out and you need to set the vst to mix in the route option if loading more than one

for Example

Guitar Rig {First “If Playing backing tracks / Drum loops this way it dont get affected by other VSTs”}
3rd Party VST { routing set to mix }
Amplitube { routing Set to mix }

You can rearrange the order of the chain using the Up and Down buttons in Settings | Effect plugins.

More info on the Route setting:

• On - the plugin processes input audio and you hear only the output of the plugin (Wet 100%)
  
• Mix - the plugin processes input audio but you hear a 50/50 mix of the plugin output and original input (Dry 50%, Wet 50%)
  
• Bypass - you hear the original input and the plugin is skipped (Dry 100%)
